Performance Evaluation of "toROMAN™" Product.
In the first performance evaluation of the toROMAN™ product, a model with Density Gradient and the most well-known microfluidic sperm sorting chip was used in the same environment. In the evaluations, concentration, motility and DNA fragmentation index results were compared. Fertility rates, clinical pregnancy and birth rates will be evaluated through ongoing studies.
toROMAN™ Usage Protocol:
- First, incubate the collected semen sample at room temperature for 15–25 minutes to allow liquefaction.
- During this period, keep the toROMAN™ device at 37ºC until use.
- Using a 100–1000 µL pipette or a 1 mL syringe, load the sperm washing solution into the outlet area according to the volumes specified in the table below. After loading, wait for 10–15 seconds to ensure that all microchannels are completely filled.
- Using a 100–1000 µL micropipette equipped with the recommended tip or a 1 mL syringe, slowly dispense the semen sample volumes indicated in the table into the inlet ports. Allow the semen sample to drip freely. Do not touch the surface with the pipette tip.

- Afterwards, the prepared toROMAN™ device is incubated for 2 hours at 37°C in a CO₂-free incubator with gentle handling.
- Following incubation, the preparation is examined under an inverted microscope, and the sperm cells reaching the full outlet area are evaluated.
7. Using a micropipette, approximately 750 µL of sorted sperm cells can be collected from the outlet area.
- The collected sperm cells are then transferred to the designated treatment areas.
